Lost Prime with water filter change

Long story but I had to remove the water separator to get the cap off. I got it back on but it seems to have lost its prime. I have tried a few times to get it started. I don't want to crank it for too long.

Am I ok to keep doing that or am I going to have to crack the fuel lines

It's a 2014 6.7

Forget it...I fixed it. Evidently the fuel line to the injection pump has to be hooked up too??
