OD and TQ are engaged by PCM based on speed reading from Trans Output Sensor. If one works and not the other, this is likely not the problem, but if they both don't engage, that's where I'd start, unless you're in limp mode.
If it's the TQ that's not engaging, common problem there is the brake switch. Try holding the peddle up with your foot while driving and see if it works then.
If those all check out, wire up a switch for the OD and/or a switch for the TQ, although you'll have to check into which wires to tap into as I'm not sure if it's the same from V10 to BigC. If you have TQ or OD (whichever isn't working) with the manual switch, you'll know you're ok mechanically and electrically up to the point where you're tapped in. If not, the problem is between the spot you hooked into the control wire and the OD/TQ piston. That's assuming you're not in limp mode as the trans relay cuts solenoid power when in limp mode.

ALWAYS Start with Ur basics im as guilty as the next for wanting to jump to something major wrong. Fluid Level and condition. a low fluid level will give you all sorts of problems. if its low time to start looking for leaks
